What effect do you suspect the following would have on your judgement of the Gram reaction of an organism?

Using an old Gram's Crystal Violet preparation containing crystalized dye.



Increasing the length of time for counterstaining.



Destaining with ethanol for one minute.



Susbstituting aqueous crystal viloet for Grams' Crystal Violet.

What effect do you suspect the following would have on your judgement of the Gram reaction of an organism?
using an old gram stain would create precipitate on your slide. if you are not well trained in reading gram stains, you might mistake them as gram positive organisms.



increasing the length of counterstaining would make the organisms pink or falsely gram neg.



destaining for one minute is too much. you would overdecolorize the bacteria and it will all turn out gram neg.



sorry, don't know the answer to last question. i've never substituted it with aqueous primary stain... i don't think it will make a big difference though.
